OVERVIEW
TRIGLUMA is a next-generation peptide introduced by JN Beauty Solutions®, belonging to the GLP-1/GIP/Glucagon peptide (triple-receptor agonist) class. It is being developed to support the management of obesity, type 2 diabetes, and other metabolic disorders.
Unlike conventional weight-loss medications, TRIGLUMA simultaneously targets multiple hormonal pathways to address the root causes of metabolic imbalance. HOW TRIGLUMA WORKS
After subcutaneous injection, TRIGLUMA activates three key receptors:

TRIGLUMA (active ingredient: Retatrutide) has been evaluated in multiple large-scale clinical trials:
- Phase 2 Trial (48 weeks): Participants with obesity receiving 12 mg weekly achieved an average weight loss of 24.2%, with continued weight reduction observed at the end of the study and no plateau reached.
- Phase 3 TRIUMPH-4 Trial (topline results, December 2025): In adults with obesity or overweight and knee osteoarthritis (without diabetes), the 12 mg dose delivered an average weight loss of 28.7% (approximately 71.2 lbs / 32.3 kg) over 68 weeks. Significant improvements in knee pain and physical function were also reported.
- Phase 3 TRANSCEND-T2D-1 Trial (topline results, March 2026): In patients with type 2 diabetes, the 12 mg dose achieved an average HbA1c reduction of up to 2.0% and mean weight loss of 16.8% (approximately 36.6 lbs / 16.6 kg) at 40 weeks. Weight loss continued without plateau through the end of the treatment period.
ADDITIONAL OBSERVED BENEFITS INCLUDE:
- Substantial reduction in visceral fat, improved lipid profiles, and lowered blood pressure.
- Enhanced fat oxidation and better preservation of lean muscle mass when combined with adequate protein intake and resistance training.

ADMINISTRATION
Dosage Form: Subcutaneous injection (once weekly)
Dosing in Trials: Starts at 1 mg per week, with gradual titration up to 12 mg per week based on tolerability and treatment goals.
COMMON SIDE EFFECTS

WHO SHOULD NOT USE TRIGLUMA?

HOW DOES TRIGLUMA COMPARE TO OTHER PEPTIDES?
Compared to Semaglutide (GLP-1 agonist only) and Tirzepatide (GLP-1 + GIP dual agonist), TRIGLUMA offers a superior triple-agonist mechanism by also activating the glucagon receptor. This not only enhances weight-loss efficacy but also increases energy expenditure and metabolic flexibility.
